Graduate student Jennifer Polinski recently traveled to California to attend the 2015 Workshop on Genome-Wide SNP Genotyping with 2bRAD at the Wrigley Marine Science Center on Catalina Island. 2bRAD genotyping uses Type IIb Restriction Endonuclease Associated DNA markers to define a subset of the genome for next generation sequencing, creating a cost-efficient method for genome-wide SNP discovery and genotyping. During the course of the nine day workshop, Dr. Eli Meyer, co-creator of the 2bRAD method, taught participants how to create 2bRAD sequencing preps for 16 samples: 6 Crassostrea gigas samples as part of a group project and 10 of their own samples. Jennifer chose to bring Montastraea cavernosa samples in order to compare these new data with data already generated through microsatellite analyses. In addition to learning sample preparation techniques, participants also learned how to process and analyze the resulting sequencing data. We can see the strengths of such an approach as 2bRAD and are hopeful that we may be able to incorporate Jennifer’s new knowledge into future projects!
